Principal Protein Scientist

CK Group are working in partnership with Avacta to recruit for a Principal Protein Scientist to join them at their state of the art facilities in Cambridge, on an initial fixed term contract of 12 months.

About the company:
Avacta is developing Affimer® bio-therapeutics to meet the growing demand for the next generation of drugs that will compete with monoclonal antibodies and other antibody mimetics, primarily in oncology. The Affimer technology is a novel protein scaffold based on the human protease inhibitor Stefin A. Key benefits of the Affimer technology are its small size, ease of formatting, high expression yields (even of large multimeric structures) and stability. Avacta are also developing a range of novel FAP-a activated pro-drug chemotherapeutics (pre|CISION? and TMAC?) that can be used as mono- or as combination therapies for the treatment of a wide range of cancers. Their first drug AVA6000, has recently entered clinical trials.

Reporting to the Head of Protein Science and managing a small team of scientists, you will be responsible for:
? Mentoring and line managing members of the protein team and approval of SOPs
? Project managing collaborations or internal programs, with knowledge of the target biology
? Cloning, expression and purification of Affimers for protein characterisation work, binding studies and stability testing using microbial and mammalian expression systems.
? Cloning, expression and purification of recombinant proteins to be used in phage screens and ELISA/cell-based assays from a range of expression platforms and fully characterising them using a range of methods, including HPLC, SDS-PAGE, western blot, ELISA, IEF and BIAcore
? Engineering Affimers for improved properties and with serum half-life extension technologies (eg, as Fc-fusions).
? Carrying out optimisation studies to increase Affimer yield
? Developing bench scale protein purification protocols using conventional chromatography resins to purify gram quantities of Affimer which are scalable
? Carrying out accelerated and real time stability studies on Affimers and characterising changes that may occur
? Establishing pre-formulation studies for increasing stability of the proteins produced
? Production of high quality Affimer and tool molecules for pre-clinical studies
? Producing and characterising mutated Affimer scaffolds to improve biophysical properties
? Presenting work internally and to external clients, and producing accurate scientific reports

To be considered for the position you should have the following skills and expertise:
? PhD level in protein biochemistry, protein engineering or molecular biology
? Significant practical experience of working in a laboratory in the pharmaceutical and/or biotech industry
? Experience with standard protein production techniques and development of multistage chromatography separations for protein purification technology transfer on scale
